The “rippled”
β
-sheet was hypothesized by Pauling and Corey in 1953, but remained an obscurity until relatively
recently. In 2017 we demonstrated that the addition of mirror-image peptide can be used to convert Amyloid
β
into fibrils with unique morphology and reduced toxicity. Subsequent cross-disciplinary research combined
experiment and theory, identifying rippled
β
-sheet formation as the underlying mechanism, yielding the first
rippled
β
-sheet X-ray crystal structures in 2022.
