Super-resolution microscopy has begun to transform biological and biomedical research by allowing researchers to observe structures well below the classic diffraction limit of light. DNA point accumulation in nanoscale topography (DNA-PAINT) enables super-resolution microscopy by employing the predictable, transient hybridization between short dye-labeled 'imager' and complementary target-bound 'docking' oligonucleotides. In this talk, I will present the usage of DNA-PAINT to resolve various cellular probes in situ, ranging from membrane associated targets to nuclear structures. In addition, I will show the employment of DNA-PAINT to obtain multi-'color' super-resolution imaging in mammalian cells.
