Leerdoelen At the end of the course, the student is able to:
describe and explain the main steps and principles of the methods discussed in the course

explain what defines a good drug and drug target
define the 3D topology of drug molecules based on their 2D chemical structures
identify protein-ligand interactions and explain their contributions to drug binding and specificity, using information from 3D structures and functional assays.
explain (and calculate) how changes in a drug, drug-target and their environment influence ligand binding
discuss the quality of a crystallographic data set and the associated protein structure (electron density, symmetry, statistical data qualifiers, structure validation)
evaluate and discuss the strengths and weaknesses of drug docking procedures
Omschrijving The course presents an overview of structural biology and structural bioinformatics, focusing on their use in drug discovery and design. Special attention will be given to the determination, assessment and analysis of 3D protein structures and protein-ligand interactions. Relevant case examples (e.g. HIV protease inhibitors, ACE inhibitors, drugs against adrenergic receptors) will be presented to illustrate how structural biology can accelerate drug discovery. Besides lectures, the course will also include interactive computer practicals/tutorials to allow a better understanding of how methods in structural biology/structural bioinformatics work and are carried out.

Entreevoorwaarden The course assumes prior basic knowledge about chemical bonding, protein structure, bioinformatics and biophysics obtained from the following relevant courses in years 1 and 2 of the Chemistry BSc programme : Molecules: Structure, Reactivity and Function (CHMOL-10); Biochemistry (CHBC-10); Spectroscopy (CHAC-10) and Recombinant DNA and Biotechnology (CHRDNAB-11)
