Abstract

In this lecture I will present different applications of fluorescence microscopy with single-molecule sensitivity. In the first part of the lecture I will show how optical "super-resolution", i.e., optical imaging with a resolution better than the diffraction limit of ~250 nm, is nowadays possible. Different experimental realizations will be discussed as well as applications in biology and nanotechnology (compare Figure). Ongoing efforts in our group here at the Zernike Institute will also be presented. In the second part of the lecture I will focus on single-molecule fluorescence microscopy as a tool to monitor unsynchronized (bio)chemical reactions in real-time. Different applications and projects in our group that range from chemistry to biology will be discussed.
